Abstract

Latar Belakang Virus Dengue serotipe 2 (DENV-2) merupakan serotipe paling banyak ditemukan sebanyak 80% dengan kasus infeksi sekunder yang dapat menyebabkan keparahan yang fatal seperti Demam Berdarah Dengue (DBD), dan Sindrom Syok Dengue (SSD). Kasus infeksi Dengue di beberapa wilayah Indonesia meningkat sejak januari 2020, dan ditengah pandemi COVID-19. Terapi hanya terdiri dari perawatan suportif, dan diperlukan perawatan khusus antivirus. Beberapa penelitian mengatakan bahwa kandungan zat kakao memiliki peran sebagai antivirus. Sehingga perlu penanggulangan virus Dengue dengan alternatif antivirus seperti kakao.
Tujuan Menguji efektiviitas ekstrak kulit biji kakao terhadap penghambatan replikasi virus dengue Serotipe 2 New Guinea C
Metode Penelitian ini merupakan penelitian eksperimental dengan desain post-test only control group design. Penelitian ini menggunakan 6 kelompok, 2 kelompok kontrol dan 4 kelompok perlakuan. Kelompok perlakuan terdiri dari pemberian ekstrak dari kulit biji kakao (Theobroma cacao L.) dengan konsentrasi bertingkat 10 μg/ml, 20 μg/ml, 40 μg/ml, 80 μg/ml, kelompok kontrol yaitu kelompok kontrol negatif pemberian Dimethyl sulfoxide (DMSO) 0,1%, sedangkan kontrol positif pemberian Cylosphorin A (CsA). Data diuji menggunakan uji Kruskal Wallis dengan uji Mann-Whitney.
Hasil Konsentrasi 10 μg/ml memiliki efektivitas daya hambat paling tinggi sebesar 54%, dan pada uji Kruskal Wallis terdapat nilai P=0,007 dan pada uji post hoc didapatkan perbedaaan daya hambat pada masing-masing konsentrasi.
Kesimpulan Ekstrak kulit biji kakao (Theobroma cacao L.) memiliki daya hambat terhadap virus dengue.

Background Dengue virus serotype 2 (DENV-2) is the most prevalent serotype of Dengue virusup to 80% with secondary infection cases can cause severity like Dengue Hemorrhagic Fever (DHF), and Dengue Shock Syndrome (SSD). Dengue infection cases in several parts of Indonesia had increased since January 2022, and in the COVID-19 pandemic. Therapy only of supportive care, and special antiviral treatment is required. Some studies said cocoa substances contains as an antiviral. So it is necessary to overcome the Dengue virus alternative antivus such as cocoa.
Aim To tested the effectiveness of cocoa seed skin extract against the replication inhibition of Dengue Virus Serotype 2 New Guinea C
Methods This study is an experimental with a post-test only control group design
research design. This study uses 6 groups, namely 2 control groups and 4
treatment groups. The treatment group consisted of extracting cocoa seed skin (Theobroma cacao L.) with a concentration of 10 μg /ml, 20 μg /ml, 40 μg /ml,
and 80 μg /ml. The control group was a negative control is giving Dimethyl
sulfoxide (DMSO) 0.1%, while positive control is giving Cylosphorin A
(CyA). Data were tested using the Kruskal Wallis Post hoc
Mann-Whitney.
Result Concentration of 10 μg /ml has the highest inhibition effectiveness of 54%, Kruskal Wallis test there were P-value= 0,007, and the Post Hoc test there were difference between concentrations.
Conclusion Cocoa seed skin extract (Theobroma cacao L.) has inhibitory power against dengue virus.
