Abstract
Latar Belakang : Diabetes Melitus Tipe 2 adalah penyakit metabolik paling sering
terlihat pada orang yang lebih tua dari 45 tahun ketika respons terhadap insulin berkurang
atau mengalami resistensi insulin. International Diabetes Federation (IDF) mencatat
463 juta orang diusia 20-79 tahun merupakan penyandang diabetes pada tahun
2019 atau sebanyak 9,3%. Genetik termasuk dalam faktor yang terlibat dalam
patofisiologi terjadinya Diabtes Melitus tipe 2. Gen TCF7L2 merupakan faktor transkripsi
yang meregulasi sel pulau pancreas. Belum banyak penelitian mengungkapkan hubungan
antara gen TCF7L2 dengan kejadian DMT2 terutama di Indonesia.
Tujuan : Mengetahui ada atau tidaknya asosiasi Polimorfisme Gen Transcription Factor
7-Like 2 (TCF7L2) rs7895340 Sebagai Faktor Risiko Kejadian Diabetes Melitus Tipe 2.
Metode : Penelitian ini merupakan penelitian observasional analitik dengan desain case
control. Dengan Teknik sampling menggunakan consecutive sampling dengan jumlah
responden 30 kasus dan 30 kontrol. Data diperoleh dengan cara ARMS PCR, dan
pemeriksaan Gula Darah. Analisis data yang dilakukan adalah analisis univariat, bivariat
(Fisher Exact Test).
Hasil : Seluruh subjek penelitian ini memiliki polimorfisme pada gen TCF7L2 dengan
frekuensi 30(100%) subjek kasus dan 30(100%) subjek kontrol. Hal tersebut terbukti
melalui nampak adanya 2 band pada visualisasi GE. Hasil analisis menunjukkan bahwa
tidak terdapat asosiasi antara polimorfisme gen TCF7L2 sebagai faktor risiko kejadian
DMT2 (p= NA; OR=NA).
Kesimpulan: Tidak terdapat hubungan antara Polimorfisme gen TCF7L2 sebagai Faktor
Risiko Kejadian Diabetes mellitus Tipe 2.

Backgrounds : Type 2 Diabetes Mellitus is a metabolic disease most commonly seen in
people older than 45 years when the response to insulin is reduced or insulin resistance is
present. The International Diabetes Federation (IDF) noted that 463 million people aged
20-79 years were people with diabetes in 2019 or 9.3%. Genetics is one of the factors
involved in the pathophysiology of type 2 diabetes mellitus. The TCF7L2 gene is a
transcription factor that regulates pancreatic islet cells. Not many studies have revealed
the relationship between the TCF7L2 gene and the incidence of DMT2, especially in
Indonesia.
Objectives : Knowing whether or not there is an association of Transcription Factor 7-
Like 2 (TCF7L2) rs7895340 Gene Polymorphisms as a Risk Factor for Type 2 Diabetes
Mellitus.
Methods : This research is an analytic observational study with a case control design.
The sampling technique used consecutive sampling with the number of respondents 30
cases and 30 controls. Data were obtained by means of ARMS PCR, and blood sugar
examination. Data analysis performed was univariate, bivariate (Fisher Exact Test)
analysis.
Result : All subjects in this study had polymorphisms in the TCF7L2 gene with a
frequency of 30 (100%) case subjects and 30 (100%) control subjects. This is evident
through the appearance of 2 bands on the GE visualization. The results of the analysis
showed that there was no Association between TCF7L2 gene polymorphisms as a risk
factor for the incidence of DMT2 (p = NA; OR = NA).
Conclusion : There is no association between TCF7L2 gene polymorphisms as a risk
factor for type 2 diabetes mellitus.
