Abstract
Latar Belakang Demam Berdarah Dengue (DBD) merupakan penyakit yang
disebabkan oleh virus dengue yang ditransmisikan melalui gigitan nyamuk Aedes
aegypti. Virus dengue memiliki empat serotipe dengan DENV-2 yang paling
dominan ditemukan di Indonesia. Algoritme penatalaksanaan dengue sampai saat
ini masih menangani gejala secara simtomatik. Beberapa penelitian menyatakan
bahwa senyawa metabolit sekunder dari daun singkong memiliki peran sebagai
antivirus.
Tujuan: Membuktikan pengaruh fraksi butanol, etil asetat, dan n-heksana daun
singkong (Manihot esculenta C) dalam menghambat replikasi virus dengue
serotipe 2 strain New Guinea C.
Metode: Penelitian ini bersifat eksperimental dengan rancangan post test only
with control group design. Penelitian ini menggunakan 15 kelompok perlakuan,
yang terdiri dari pemberian fraksi butanol, etil asetat, dan n-heksana daun
singkong (Manihot esculenta C.) dengan konsentrasi 20 µg/ml, 10 µg/ml, 5
µg/ml, 2,5 µg/ml, dan 1,25 µg/ml.
Hasil: Pada uji Kruskal Wallis terdapat perbedaan daya hambat antar fraksi
terhadap penghambatan replikasi virus dengue (nilai P = <0,000). Nilai IC50 fraksi
butanol 2,7 µg/mL, etil asetat 0,2 µg/mL, dan n-heksana 2,0 µg/mL menunjukkan
ketiga fraksi memiliki daya hambat dan tidak menunjukkan adanya sitotoksisitas
terhadap sel vero dengan nilai selectivity index (SI) fraksi butanol >7, etil asetat
>100, dan n-heksana >10
Kesimpulan: Fraksi butanol, etil asetat, dan n-heksana daun singkong (Manihot
esculenta C.) memiliki pengaruh dalam menghambat replikasi virus dengue.

Background: Dengue Hemorrhagic Fever (DHF) is a disease caused by dengue
virus, transmitted through Aedes aegypti mosquito’s bites. There are four different
dengue virus serotypes, with DENV-2 being the most prevalent in Indonesia. The
dengue management algorithm, as of now, is still focusing on symptomatic
treatment. Some studies showed that secondary metabolites from cassava leaves
have an antiviral function.
Aim: To demonstrate the effect of butanol, ethyl acetate, and n-hexane fraction of
cassava leaves (Manihot esculenta C) in inhibiting dengue virus serotype 2 strain
New Guinea C replication
Methods: This study used an experimental post-test only with control group
design. The 15 treatment groups in this investigation each contained fractions of
cassava leaves (Manihot esculenta C.) in butanol, ethyl acetate, and n-hexane at
concentrations of 20 µg/ml, 10 µg/ml, 5 µg/ml, 2.5 µg/ml, and 1.25 µg/ml.
Results: Kruskal Wallis test indicated differences in the inhibitory power among
fractions towards dengue virus replication (P value = 0.000). The butanol
fraction with IC50 value of 2.7 µg/mL, ethyl acetate of 0.2 µg/mL, and n-hexane
of 2.0 µg/mL exhibited inhibitory power and did not exhibit any cytotoxicity to
vero cells with selectivity index (SI) values of n-hexane >10, ethyl acetate >100,
and butanol fraction >7
Conclusion: Butanol, ethyl acetate, and n-hexane fractions of cassava leaves
(Manihot esculenta C.) had an effect in slowing down dengue virus replication.
