Abstract
Latar Belakang: Obesitas adalah penyakit multifaktorial, faktor penyebab terjadinya obesitas adalah ketidak seimbangan antara jumlah kalori yang masuk dan dikeluarkan. Mengkonsumsi makanan tinggi kalori seperti fast food dapat menyebabkan ketidakseimbangan energi yang dapat menyebabkan obesitas selain faktor lingkungan faktor genetik juga berperan dalam terjadinya obesitas salah satunya dari polimorfisme gen FTO rs9939609 .Beberapa penelitian melaporkan adanya hubungan antara polimorfisme gen FTO rs9939609 dan frekuensi konsumsi fast food terhadap kejadian obesitas.
Tujuan: Penelitian ini bertujuan untuk mengtahui hubungan polimorfisme FTO rs9939609 dan frekuensi konsumsi fast food terhadap kejadian obesitas pada dewasa.
Metode: Penelitian observasional analitik dengan desain case control yang dilakukan di Laboratorium Biomolekuler dan Genetika Fakultas Kedokteran UGJ melibatkan 84 orang dengan masing-masing kelompok kasus 42 orang dan kelompok kontrol 42 orang. Data diperoleh dengan menyebarkan formulir pendaftaran dan pengukuran frekuensi konsumsi fast food dengan instrument kuisioner, dan genotyping menggunakan PCR-RFLP, dan visualisasi menggunakan gel elektroforesis 2,5%. Analisis data dilakukan dengan chi square.
Hasil: Hasil penelitian ini menunjukkan bahwa tidak terdapat hubungan yang signifikan antara polimorfisme FTO rs9939609 terhadap kejadian obesitas p>0,05 dengan OR: 0,710, namun pada variable pola makan frekuensi konsumsi fast food terdapat hubungan signifikan p<0,05.
Kesimpulan: Obesitas dapat terjadi karena ketidakseimbangan antara energi yang masuk dan dikeluarkan, faktor kebiasaan menjadi faktor resiko yang paling berpengaruh terhadap kejadian obesitas pada penelitian ini.

Background: Obesity is a multifactorial disease, the contributing factor to the occurrence of obesity is an imbalance between the number of calories entered and expended. Consuming high-calorie foods such as fast food can cause energy imbalances that can cause obesity in addition to environmental factors genetic factors also play a role in the occurrence of obesity is one of them from the polymorphism of the FTO gene rs 9939609 . Several studies have reported an association between the polymorphism of the FTO gene rs9939609 and the diet of the frequency of fast food consumption to the incidence of obesity.
Objective: This study aims to determine the relationship of FTO polymorphism rs9939609 and the frequency of fast food consumption to the incidence of obesity in adults.
Method: Analytical observational research with case control design conducted at the Biomolecular and Genetics Laboratory of the UGJ Faculty of Medicine involved 84 people with each group of cases of 42 people and control group of 42 people. Data were obtained by disseminating registration forms and measuring the frequency of consumption of fast food with questionnaire instruments, and genotyping using PCR-RFLP, and visualization using electrophoresis gels 2,5%. Data analysis was performed with chi square.
Results: The results of this study showed that there was no significant association between FTO polymorphisms in rs9939609 on the incidence of obesity p>0.05 with OR: 0.710 CI 95%, but in dietary variables frequency fast food consumption has a significant relationship of p<0.05.
Conclusion: Obesity can occur due to an imbalance between incoming and expended energy, environmental factors are the risk factors that most influence the incidence of obesity in research this.
